I’ll say George Clooney and Michelle Pfeiffer in One Fine Day. The tension builds between them and you really believe they’re desperate for each other at the end. Probably his best acting role ever.
I have seen a lot of George Clooney movies and I do not recall him having chemistry with anyone. Seriously. But the hottest straight chemistry I can recall and maybe it's because I was younger, was between Antonio Banderas and Anjelina Jolie in Original Sin.

Well, we know why now, but the chemistry between Brad Pitt and Angelina Jolie in Mr. and Mrs. Smith made the movie fun, in spite of it being somewhat boring otherwise. We later found out they were having an affair while filming, so that probably explains that great chemistry.

Clooney has zero heat with women on and off screen. When he was younger his good looks deflected from the lack of chemistry, now it's glaring obvious. He and JLo couldn't stand each other, so it was a Richard Gere/Debra Winger situation where their contempt for one another came off as chemistry.

I watched "Accidental Tourist" with a friend. Neither of us had read the book or knew the plot. In the middle of the movie, my friend asked me: Who do you think he'll end up with? I said Muriel (Geena Davis' character). It just made sense to me and made sense for the plot that he would end up with someone who actually seemed to need him more. (The Kathleen Turner character was independent and Hurt's character needed to be needed.)
