Are we talking total profit or a percentage based on cost ratio? If we are talking cost ratio I would say some small Indy movie that became a cult film like "The Toxic Avenger" or something like that.

Yes you’d have to clarify if you’re adjusting for inflation and including budge/cost/marketing and all that.
I’ve heard conflicting reports over the years. Gone With the Wind is usually considered the most profitable in sheer box office dollars when adjusted for inflation. But it also cost a tremendous amount of money to make in 1930’s dollars. Same with Titanic, which at a cost of 200 million to make, was the most expensive movie ever made up to that time.
Deep Throat-of all movies!- has been listed as a candidate because it cost mere thousands to make but turned profits in the hundreds of millions.

Reports started coming out in the 80’s about how few Hollywood “blockbusters” actually turn a significant profit. One of the film magazines did a breakdown of an Eddie Murphy movie that was being touted as a big success at the time. By time they took into account of how much was budgeted on marketing, star salaries, and perks etc, it was revealed that the movie still hadn’t turned a profit a couple years after it’s release. I think it was Coming To America.
It was revealed how important overseas sales, TV rights, video sales etc was to a Hollywood movie actually making money for the studios.
