Firstly, I don't think a movie is long enough to establish anything more than a fuck session, which is what most of these suggestions are wishing for, and that's of little interest to me.
I want to see a relationship - a smart, funny, sexy relationship - and it doesn't have to involve anyone I would actually want to get with myself, as long as the actors are able to portray people that seem to be legitimately into each other, and the relationship is given enough time to play out, so: television.
That being said, and I know this is WAY out of left field, but...
Dennis Boutsekaris & Gary Cole
I know, I know - WTF?!
BUT, they had a scene opposite each other in an episode of The Good Wife where Dennis (probably playing a lawyer) was grilling Dennis, who plays a ballistics expert, and I remember thinking, "Huh. That's interesting."
Both attractive in their own ways, fiercely intelligent, both have dramatic chops with good senses of humor - could play men from different religious, socio-economic backgrounds. I'd watch that!
Their connection made enough of an impact that I jotted the episode down.

The kiss between Josh Hartnett and Reeve Carney is one of the most intense scenes I have ever scene on television. I mean the build up is breathtakingly sinister and passionate at the same time. When Josh walks up to Reeve for the kiss. I almost fainted.
These two need to be in a film together playing a couple.

Absolutely, R30, one of the most intense moments I've ever seen on TV as well-- something I thought could have happened from earlier in the episode, but it still literally took my breath away. The sumptuous design of the series in general didn't hurt matters, as well as the respective actors' handsome looks. I want more with both of them, dammit!

Unconventional as it sounds, based on their chemistry in NIKITA I’d like to see Sawa & Aaron Stanford play a gay couple.
I can see them making a magical-realist black comedy where they play two delusional drifting addict-romantics in love, who believe that they can find refuge and set up a safe gay ‘Paradise’ deep in the desert where an increasingly anti-gay Administration can’t reach. Stanford could be an impulsive dreamer with a bold flamboyance who plunges into his narcotic adventure with an air of defiance. Sawa could play the quieter & more submissive of the two, who follows Stanford’s vision with a hopeful desperation to stay together and live free of the fears inside his mind.
I imagine that even though they’re paranoid tinhat types with wild imaginations and a very insecure lifestyle, these two lovers are wideyed & well-intentioned in spite of that and genuinely share a tender passion for one another (and of course there will be at least one extended lovescene with no discretion shots, to unsettle the straights).
It could have musical numbers, and fantasy sequences with plenty of psychedelic weirding; think YELLOW SUBMARINE & PRISCILLA: QUEEN OF THE DESERT meeting THE LIVING END in tone & style. I would also want plenty of cameos from DL faves and a liberal sprinkling of our in-jokes included as Easter eggs, just for all of you.
